Protein function regulation through inserts for response to biological, chemical and physical signals
Principal investigator: prof. dr. Roman Jerala (ERC Advanced Grant)
The vision of the project is to accelerate the introduction of new properties into proteins as key molecules in all biological systems. Rational design of responses to different types of signals will provide insight into the functioning of biological systems and the engineering of biological systems.
The project draws on the INSRTR platform for allosteric regulation of protein function (Plaper et al., 2024, Cell Discovery):
Objectives:
1. Design new protein regulators and regulators of natural proteins
2. Engineering of modular protein responsiveness to pH and RNA, to design proteins that will sense and regulate protein function depending on specific RNA molecules.
3. Design of proteins sensing other proteins and construction of interacting protein networks for concentration-dependence of protein function and biomedical applications
4. Construct new mechano-responsive proteins, genetically encoded sensors for a wide range of mechanical forces
