Four-helix bundle segmentation strategy

Designed interactions between proteins are an excellent tool for the development of diverse applications in synthetic biology and biomedicine. With the desire to expand the possibility of oligomerization, we introduced a segmentation strategy of four-helix bundles (4HB), with which we separated 4HB into different parts (1:3, 2:2, 1:2:1, 1:1:1:1), which enable the assembly of up to four different proteins into a protein complex. The prepared oligomerization modules were implemented in the regulation of mammalian cells for the assembly of split enzymes, gene expression, or regulation of CAR-T cells.
